AN EFFICIENT HPLC-UV METHOD FOR DETERMINATION OF TETRAHYDROCANNABINOL IN OIL

Objective: The purpose of this work was to develop and validate a high-performance liquid chromatography (HPLC) with ultra-violet (UV) detection method for the determination Δ9-tetrahydrocannabinol (THC) in oil. Methods: HPLC analysis carried out using an Agilent 1260 Infinity Series® II system detection. stationary phase consisted ACE 5 μm C18 column (250×4.6 mm) at temperature 40°C. mobile acetonitrile phosphate buffer (pH=2.5) (70:30, v/v) flow rate 2 mL/min. injection volume 20 μL. UV detector set 220 nm. Separation THC from oil outusing solvent extraction: 0.6 mL methanol, 0.3 two drops internal standard were placed centrifuge tube, vortex mixed 30 s, sonicated min, centrifuged 6000 rpm 15 min. organic layer separated, stored −20°C h, separated again, passed through syringe filters 0.45 μm, analyzed. Results: found be linear range 0.039–5.000 μg/mL, acceptable inter- intra-assay precision, accuracy, stability. limit 0.019 μg/mL quantification 0.039 μg/mL. Conclusion: developed is quick easy use can determine good peak shape resolution. Application analytical will help CBD medicinal use.
